Structural and spectroscopic characterization of a HdrA-like subunit fromHyphomicrobium denitrificans

Summary: Many bacteria and archaea utilize a novel pathway of sulfur oxidation similar to that of methanogens, involving a key role of heterodisulfide reductase (Hdr) in the reaction. The structural analysis of Hdr-like proteins from sulfur oxidizers revealed similarities and differences with methanogenic Hdr proteins in terms of electron transfer and reaction mechanisms. The findings suggest a potential mechanism linking protein-bound sulfane oxidation to sulfite in sulfur oxidizers.

Summary: In methanogenic archaea, the HdrABC and MvhAGD complexes couple the reduction of CO2 and heterodisulfide via flavin-based electron bifurcation. The modular architecture of HdrA(BC) enzymes allows for functional diversity, but questions regarding electron transfer and energy coupling processes remain. Despite known crystal structures, fundamental aspects of HdrA(BC) core modules integrated into electron-transfer machineries are still being explored.
