Summary: The chemotherapy drug paclitaxel (PTX) can increase expression of sodium channel Na(v)1.7 in sensory neurons, leading to enhanced excitability. This study utilized live imaging to show concentration and time-dependent effects of PTX on vesicular trafficking and membrane localization of Na(v)1.7 in sensory axons. PTX treatment increases functional Na(v)1.7 channels in dorsal root ganglion neurons and enhances trafficking and surface distribution in sensory axons, with outcomes influenced by the presence of an inflammatory milieu, potentially explaining increased excitability and pain in CIPN.
