Meta-Analytical Accuracy of ANCA Renal Risk Score for Prediction of Renal Outcome in Patients With ANCA-Associated Glomerulonephritis

Mengdi Xia et al.

Summary: This meta-analysis highlighted the good performance of the ARRS score in predicting renal outcomes in patients with ANCA-GN. The cumulative rate of ESRD increased with higher ARRS categories. The sensitivity and specificity of ARRS & GE; 2 and ARRS & GE; 8 differed, with the former having higher sensitivity and the latter having higher specificity.

Evaluation of a newly proposed renal risk score for Japanese patients with ANCA-associated glomerulonephritis

Masaya Saito et al.

Summary: This study is the first to report the usefulness of the renal risk score (RRS) for predicting renal outcomes among Japanese patients with antineutrophil cytoplasmic antibody-associated glomerulonephritis (AAGN). The predictive value of RRS was comparable to that of conventional prognostic tools.

Assessment of Renal Risk Score and Histopathological Classification for Prediction of End-Stage Kidney Disease and Factors Associated With Change in eGFR After ANCA-Glomerulonephritis Diagnosis

Benoit Brilland et al.

Summary: This study analyzed the value of Renal Risk Score (RRS) and histopathological classification in predicting the risk of end-stage kidney disease (ESKD) in ANCA-associated glomerulonephritis (ANCA-GN). It also found that eGFR at ANCA-GN diagnosis was the main determinant of eGFR variation within the first 2 years.

New Therapeutic Targets in Antineutrophil Cytoplasm Antibody-Associated Vasculitis

Maria Prendecki et al.

Summary: ANCA-associated vasculitis is a rare systemic autoimmune disease characterized by inflammation of small blood vessels and the presence of circulating ANCAs. Current treatment strategies involve inducing remission with high-dose glucocorticoids and either rituximab or cyclophosphamide, followed by sustained low-dose treatment to prevent relapse. New therapeutic targets are being explored to improve outcomes and reduce treatment-related side effects in AAV patients.

Summary: The study validated the renal risk score in a large cohort of southern European patients, demonstrating its accuracy in predicting renal outcomes among patients with renal vasculitis. The risk scoring system can assist in developing tailored prevention and treatment strategies for patients in different risk groups.

Development and testing of an artificial intelligence tool for predicting end-stage kidney disease in patients with immunoglobulin A nephropathy

Francesco Paolo Schena et al.

Summary: The study developed an artificial neural network prediction model for ESKD in IgAN patients, showing high performance values and recall rates over five and ten-year follow-ups. The model demonstrated successful results in an independent cohort and outperformed other mathematical models in discriminant indexes and calibration. The dynamic discrimination indexes of the artificial neural network remained consistent over a 25-year follow-up period, indicating its potential for accurately identifying high-risk individuals and predicting time-to-event endpoints.

AI applications in renal pathology

Yuankai Huo et al.

Summary: The explosive growth of AI technologies, particularly in deep learning, has led to revolutionary applications in AI-assisted healthcare, including in renal pathology. However, successful integration of AI in renal pathology requires close interdisciplinary collaborations between computer scientists and renal pathologists. Understanding the high-level principles of AI technologies and optimizing AI techniques for renal pathology are crucial for future applications in this field.

Avacopan for the Treatment of ANCA-Associated Vasculitis

David R. W. Jayne et al.

Summary: In this study, the C5a receptor inhibitor avacopan was found to be noninferior to prednisone taper in achieving remission at week 26, but superior in achieving sustained remission at week 52 in patients with ANCA-associated vasculitis. All patients received cyclophosphamide or rituximab. The safety and clinical effects of avacopan beyond 52 weeks were not addressed in the trial.

Reappraisal of Renal Arteritis in ANCA-associated Vasculitis: Clinical Characteristics, Pathology, and Outcome

Idris Boudhabhay et al.

Summary: In patients with AAV and renal involvement, arteritis of small kidney arteries is associated with worse outcomes and has specific clinical and histologic characteristics. The presence of arteritis independently predicts end-stage kidney disease and significantly improves the discriminative ability of the ANCA renal risk score. Confirmation of these findings in external validation cohorts is necessary to further establish the prognostic contribution of arteritis status.
