Bioengineering of fibroblast-conditioned polycaprolactone/gelatin electrospun scaffold for skin tissue engineering

Summary: In this study, a composite of polycaprolactone and gelatin was surface modified with gelatin coating or plasma treatment, and then cultured with fibroblast 3T3 and decellularized to fabricate a fibroblast ECM-conditioned scaffold. The results showed that the plasma-treated scaffold had better cell attachment and ECM secretion, and the conditioned scaffold exhibited improved biocompatibility and angiogenic properties in vivo.

Extracellular vesicles from three dimensional culture of human placental mesenchymal stem cells ameliorated renal ischemia/reperfusion injury

Summary: This study demonstrated that extracellular vesicles (EVs) from 3D culture of human placental MSCs (hPMSCs) showed greater therapeutic potential for ischemia-reperfusion acute kidney injury (I/R-AKI) compared to EVs from 2D culture. The EVs from 3D culture were more effective in protecting against cell death and inflammation, leading to reduced tissue damage and improved renal function. MicroRNA profiling analysis revealed significant changes in EVs from 3D culture, particularly in the expression of miR-93-5p.

Summary: This study developed a drug delivery system based on LDH and Gal for targeted delivery of Cur, showing promising results including high cytotoxicity and apoptosis effects on HepG2 cells. The Gal-Cur/LDH nanoparticles demonstrated a specific targeting efficiency due to higher cellular uptake via ASGP-receptors. Our findings suggest the Gal-Cur/LDH nanoparticles as an excellent carrier system for targeted delivery of Cur to hepatocellular carcinoma cells.

Long-term preservation effects on biological properties of acellular placental sponge patches

Summary: Decellularization, preservation protocol, and storage time affect the biomechanical and biological properties of allografts and xenografts. The decellularized placental sponge (DPS) showed decreased antibacterial and angiogenic properties over time, with three months of preservation at -20 degrees C recommended for optimal retention of these properties. This storage protocol may be beneficial for preserving similar decellularized placenta-derived products while maintaining their biological properties.

Optimization of decellularized human placental macroporous scaffolds for spermatogonial stem cells homing

Summary: The decellularized scaffold with 0.5%/30 SDS/Triton was found to have minimal negative effects on ECM, making it an excellent platform for tissue engineering applications. Different concentrations and incubation times had varying effects on swelling ratios and cell viability. The colony formation of spermatogonial stem cells was clearly observed within the scaffold.

Summary: This study demonstrates that pepsin-solubilized DCM (PDCM) can activate bone marrow-derived macrophages to polarize towards a beneficial macrophage phenotype. These polarized macrophages promote bone marrow-derived mesenchymal stem cell invasion, migration, proliferation, and chondrogenic differentiation. Combining DCM scaffolds with early intra-articular injection of IL-4 creates a wound-healing microenvironment that enhances cartilage regeneration in a rat knee osteochondral defect model.

Comparison between placental and skeletal muscle ECM: in vivo implantation

Summary: This study compared the regenerative effects of decellularized canine placentas and murine skeletal muscles in rat skeletal muscles. Results showed a higher percentage of proliferative cells in placental matrices compared to muscle matrices, with placental matrices not completely degraded at day 15. Both matrices were eventually resorbed, with morphologically normal myofibers observed after 45 days.
