Endothelial Transcytosis in Acute Lung Injury: Emerging Mechanisms and Therapeutic Approaches

Summary: Acute Lung Injury (ALI) is a severe condition that leads to respiratory compromise and death in many patients. The inflammation and damage caused by the loss of endothelial barrier integrity, pneumocyte necrosis, and leukocyte recruitment contribute to the progression of ALI. In addition, pulmonary microvascular damage by bacteria or viruses further impairs lung function. Transcytosis, the transport of protein through caveolar vesicles, has been found to occur in the early phase of ALI and may play a role in the disease process. This article discusses the role of transcytosis in healthy and injured endothelium and the potential use of caveolar transport for delivering therapeutics to the lungs to prevent further injury or improve recovery.

PLCβ2 Promotes VEGF-Induced Vascular Permeability

Summary: This study identifies PLC beta 2 as a key regulator of vascular permeability induced by VEGF, functioning through regulation of calcium flux and phosphatidylinositol 4,5-bisphosphate levels. Targeting PLC beta 2 could provide a novel approach to regulate vascular permeability in patients.

Structure and regulation of phospholipase Cβ and ε at the membrane

Summary: Phospholipase C (PLC) beta and epsilon enzymes hydrolyze phosphatidylinositol (PI) lipids in response to interactions with G protein subunits and small GTPases, generating second messengers that regulate intracellular Ca2+ concentration and activate PKC to affect physiological processes. The enzymes share a conserved core for lipase activity but utilize different strategies and structural elements to regulate their activity and interactions with membranes and G protein regulators.
