Endothelial cell-derived tetrahydrobiopterin prevents aortic valve calcification

Zongtao Liu et al.

Summary: This study found that the levels of BH4 in the circulation and aortic valve tissue were significantly decreased in patients with aortic valve calcification. Endothelial cell-specific deficiency of GCH1 contributed to the development of aortic valve calcification. Oxidized LDL induced osteoblastic differentiation of valvular interstitial cells, which was inhibited by BH4 supplementation. Generation of peroxynitrite and tyrosine nitration of DRP1 contributed to aortic valve calcification. Folic acid attenuated hypercholesterolemia-induced aortic valve calcification by increasing DHFR levels.

Aspirin relieves the calcification of aortic smooth muscle cells by enhancing the heat shock response

Quanquan Shen et al.

Summary: Aspirin can partially alleviate vascular smooth muscle cell (VSMC) calcification by regulating the expression of heat shock protein 70 (HSP70) and heat shock protein 90 (HSP90). Blocking HSP70 or HSP90 inhibitors can partially weaken the anti-calcification effect of aspirin.

Brain-Derived Neurotrophic Factor Expression and Signaling in Different Perivascular Adipose Tissue Depots of Patients With Coronary Artery Disease

Sarah Zierold et al.

Summary: In patients with coronary atherosclerosis, elevated BDNF expression and decreased TrkB receptor levels in perivascular fat suggest a reduced vascular BDNF signaling pathway. This study also found increased levels of negative regulators of neurotrophin signaling in perivascular fat, which may contribute to the loss of vascular BDNF signaling.

Red Blood Cell and Endothelial eNOS Independently Regulate Circulating Nitric Oxide Metabolites and Blood Pressure

Francesca Leo et al.

Summary: This study demonstrates that eNOS in ECs and RBCs independently contribute to blood pressure homeostasis. EC eNOS KO mice exhibited significantly impaired aortic dilatory responses to acetylcholine, loss of flow-mediated dilation, and increased blood pressure. On the other hand, RBC eNOS KO mice showed no alterations in acetylcholine-mediated dilation or flow-mediated dilation but were hypertensive.
