Redefining the coenzyme A transferase superfamily with a large set of manually annotated proteins

Summary: Coenzyme A (CoA) transferases are a diverse group of proteins involved in the metabolism of acetyl-CoA and other CoA thioesters. They catalyze over 100 biochemical reactions and span all three domains of life. Previous classification into three families is incorrect, as our analysis of characterized CoA transferases reveals different family relationships. Our manual annotation of 94 CoA transferases provides comprehensive functional information, surpassing information available in protein databases. By regrouping the families into six, including one family with non-CoA transferases, we resolve issues in sequence, structure, and catalyzed reactions. Our work is crucial for understanding CoA transferase biology and emphasizes the need to look beyond database information in phylogenetic analyses of proteins.

Aminoacylation, a rarely observed tailoring step in terpene biosynthesis, is found to be involved in IDT biosynthesis through the unusual participation of monomodular nonribosomal peptide synthetase-like enzymes.

Nodulisporic acid produces direct activation and positive allosteric modulation of AVR-14B, a glutamate-gated chloride channel from adult Brugia malayi

Summary: In this study, a GluCls subunit from Brugia malayi was cloned and its pharmacological properties were investigated. The results showed that the receptor was regulated by the natural ligand L-glutamate and ivermectin, and had dual effects when exposed to nodulisporic acid. Furthermore, the unique pharmacological characteristics of this GluCls subunit could explain the variation in drug sensitivities observed in different nematode parasites.
