The identification and application of a robust ω-transaminase with high tolerance towards substrates and isopropylamine from a directed soil metagenome

Youyu Xie et al.

Summary: This study identified a robust omega-transaminase, ATA1012, with high industrial potential from a metagenomic library. ATA1012 exhibited stability, tolerance towards isopropylamine (IPA) and high substrate tolerance, enabling efficient production of chiral amines. Through systematic optimization, the catalytic efficiency of ATA1012 was further improved. This research has significant industrial application value in synthesizing chiral amine pharmaceuticals and precursors.

Nedd8-Activating Enzyme Is a Druggable Host Dependency Factor of Human and Mouse Cytomegalovirus

Yulia Alejandra Flores-Martinez et al.

Summary: Human cytomegalovirus manipulates the proteome of infected cells through the ubiquitin proteasome pathway, with the Nedd8-activating enzyme (NAE) being indispensable for neddylation. The drug MLN4924 shows potent antiviral activity against cytomegaloviruses by inactivating CRL, but may have off-target effects due to its nucleoside analog nature. NAE, the neddylation process, and CRLs are potential druggable host dependency factors (HDFs) for cytomegaloviruses.

Computational Redesign of an ω-Transaminase from Pseudomonas jessenii for Asymmetric Synthesis of Enantiopure Bulky Amines

Qinglong Meng et al.

Summary: The study utilized computational protein engineering to expand the substrate scope of the (S)-selective omega-TA from Pseudomonas jessenii for the synthesis of bulky amines. Multiple small variant libraries were designed computationally, with the majority of variants showing desired activity in laboratory tests. The results demonstrate that computational design can accurately predict the substrate scope and enantioselectivity of PjTA mutants, enabling the rapid production of enantiopure bulky amines.
