Summary: Mass spectrometry imaging is a hot research field that can reflect the distribution of multiple metabolites in tissue. However, not all types of metabolites have high ionization efficiency in mass spectrometry imaging. This study developed a platform that combines online quaternized derivatization and laser ablation carbon fiber ionization mass spectrometry imaging, achieving significant increase in mass signals of monoglycerides and diglycerides, and readily acquiring high-quality mass images of these metabolites.

Summary: With the development of mass spectrometry imaging, a new method for absolute quantitation of endogenous compounds in bio-samples using single probe for ambient liquid extraction has been developed. By establishing a theoretical kinetic model, the new method allows for accurate per-pixel quantitation and reproducibility of lipid imaging with better contrast and wider linear range compared to conventional quantitative MSI methods. The absolute quantitation results were also verified by quantitative LC-MS method.
