Cancer therapy-related cardiac dysfunction: is endothelial dysfunction at the heart of the matter?

Crizza Ching et al.

Summary: The significant improvements in cancer survival have led to the recognition of unintended adverse cardiovascular effects associated with cancer treatment, particularly CTRCD. Emerging biomarkers such as circulating inflammatory markers, cardiac biomarkers, microRNAs, and EVs are now considered as early indicators of cardiovascular injury, while the role of endothelial cells in regulating cardiac function in CTRCD is being increasingly understood.

Damage to cardiac vasculature may be associated with breast cancer treatment-induced cardiotoxicity

Rebecca K. Hoffman et al.

Summary: Using doxorubicin and trastuzumab in an orthotopic mouse model of human breast cancer resulted in cardiac vascular defects, preserved left ventricular ejection fraction, indicating mild cardiac dysfunction. The cardiac endothelium was found to be more sensitive to doxorubicin therapy compared to endothelial cells in other organs, suggesting a correlation between cardiac endothelial damage and breast cancer treatment-induced cardiotoxicity.
