Clinical determinants of long-term survival in metastatic uveal melanoma

Elias A. T. Koch et al.

Summary: The study identified that a good response to immune checkpoint blockade, radiation therapy, or liver-directed treatments were associated with prolonged overall survival. Conversely, a poor ECOG performance status and elevated serum LDH were linked to a poorer overall survival. Long-term survivors often had more organs affected by metastasis, while patients with liver metastases only were more common among short-term survivors.

Metastatic uveal melanoma: The final frontier

Elina S. Rantala et al.

Summary: Treatment of primary intraocular uveal melanoma has advanced significantly, but the risk of distant metastases remains high. There is no consensus on surveillance, staging, and treatment of disseminated disease, making it difficult to improve survival rates. Solving these issues is the key to curing metastatic uveal melanoma.

Identification of Early-Onset Metastasis in SF3B1 Mutated Uveal Melanoma

Wojtek Drabarek et al.

Summary: This study describes the clinical and genetic characteristics of a cohort of 146 SF3B1-mutated UM, finding that SF3B1-mutated UM exhibit early- and late-onset metastatic disease without distinguishing biomarkers, with largest basal tumor diameter possibly associated with early metastasis. Analysis of gene expression and aberrant transcripts revealed an association between ABHD6 expression and early-onset metastatic SF3B1, providing potential prognostic indicators and therapeutic targets for SF3B1-mutated UM.

Genomic Profiling of Metastatic Uveal Melanoma Shows Frequent Coexisting BAP1 or SF3B1 and GNAQ/GNA11 Mutations and Correlation With Prognosis

Alexandra L. Isaacson et al.

Summary: This study aimed to identify therapeutic targets and correlate with clinical outcomes in metastatic UM through mutation profiling using NGS. The results showed that GNAQ/11-SF3B1 mutations were associated with a longer time to first metastasis compared to GNAQ/11-BAP1 mutations. Coexisting BAP1/SF3B1 and GNAQ/11 mutations were unique to UM, and SF3B1 mutations may play a role in UM metastasis.

Impact of staging on survival outcomes: a nationwide real-world cohort study of metastatic uveal melanoma

Elina S. Rantala et al.

Summary: This study analyzed the overall survival of patients with metastatic uveal melanoma and found that surgical resection can prolong survival. Staging of patients is crucial when comparing survival outcomes, and it is recommended to compare novel treatments against available BSC data for specific stages.

Multiregional genetic evolution of metastatic uveal melanoma

Daniel A. Rodriguez et al.

Summary: The study identified temporal and anatomical complexity in the genetic evolution of metastatic uveal melanoma, including multiple seeding events and the occurrence of late driver variants. This highlights the distinction between early and late phases of UM genetic evolution and has implications for novel therapeutic approaches.
