Imaging Features with Histopathologic Correlation of CNS High-Grade Neuroepithelial Tumors with a BCOR Internal Tandem Duplication

L. Cardoen et al.

Summary: A new brain tumor entity occurring in early childhood characterized by BCL6 corepressor gene internal tandem duplication presents as large intra-axial peripheral masses with well-defined borders, no edema, restricted diffusion, weak contrast enhancement, central necrosis, hemorrhage and calcifications. Intratumoral veins are observed, and there is no leptomeningeal dissemination at the time of diagnosis, providing valuable insights for histologic and genomic profiling in young children with brain tumors.

Detailed Clinical and Histopathological Description of 8 Cases of Molecularly Defined CNS Neuroblastomas

Till Holsten et al.

Summary: The study describes CNS NB FOXR2 tumors as a class of brain tumors with common genetic events and DNA methylation profiles. Histomorphological and immunohistochemical features were described, along with MRI findings for some cases. The study suggests that these tumors may fit well into the WHO entity of CNS neuroblastoma, providing helpful information for establishing an integrated diagnosis.

Molecular analysis of pediatric CNS-PNET revealed nosologic heterogeneity and potent diagnostic markers for CNS neuroblastoma with FOXR2-activation

Andrey Korshunov et al.

Summary: This study identified molecular markers, namely SOX10 and ANKRD55, as key genes distinguishing CNS_NBL from other tumors exhibiting CNS-PNET. Immunohistochemical detection of combined expression of SOX10 and ANKRD55 accurately identifies CNS_NBL, a distinct pediatric brain tumor entity, from morphological mimics. Prospective patient series are needed to confirm the diagnostic IHC algorithm due to the rarity of CNS_NBL.

Clinical phenotypes and prognostic features of embryonal tumours with multi-layered rosettes: a Rare Brain Tumor Registry study

Sara Khan et al.

Summary: Embryonal tumours with multi-layered rosettes (ETMRs) are rare paediatric brain tumours with alterations of the C19MC microRNA locus. Survival of ETMR patients is associated with non-metastatic disease, non-brainstem location, gross total resection, high-dose chemotherapy, and radiotherapy. Tailored post-surgical multi-modal therapy based on patient-specific risk features could improve ETMR survival.

Therapeutic implications of improved molecular diagnostics for rare CNS embryonal tumor entities: results of an international, retrospective study

Katja von Hoff et al.

Summary: After molecular classification of CNS-PNET based on DNA methylation analysis, the treatment outcomes of patients become highly variable. Patients with CNS NB-FOXR2 responded well to current treatments, while those with ETMR had the worst outcomes.
