Iodinated cyanine dye-based nanosystem for synergistic phototherapy and hypoxia-activated bioreductive therapy

Summary: Photodynamic therapy (PDT) combined with hypoxia-activated chemotherapy is a novel strategy to enhance anti-cancer therapy. In this study, a novel liposome was developed for PDT-activated chemotherapy, which showed improved efficacy compared to traditional PDT or chemotherapy. These liposomes based on iodinated cyanine dyes provide a powerful and versatile theranostic strategy for tumor target phototherapy and PDT-induced chemotherapy.

Conquering the Hypoxia Limitation for Photodynamic Therapy

Summary: Photodynamic therapy (PDT) is a promising treatment modality due to its high spatiotemporal selectivity, minimal invasiveness, and low systemic toxicity, however, the hypoxic nature of many solid tumors limits its therapeutic effect. Efforts have been made to increase tumor oxygen content and alleviate tumor hypoxia through various strategies developed in the past decade, including delivering exogenous oxygen, generating oxygen in situ, reducing tumor cellular oxygen consumption, regulating the tumor microenvironment, and inhibiting the hypoxia-inducible factor 1 signaling pathway.

Summary: Chemodynamic therapy (CDT) is a promising method for killing cancer cells by converting intracellular hydrogen peroxide (H2O2) into highly toxic hydroxyl radicals ((OH)-O-center dot). An intelligent nanocatalytic theranostics, PGC-DOX, was developed to address the limitations of endogenous H2O2 and high GSH levels in tumor cells, leading to efficient cancer therapy. The system combines H2O2 self-supply, GSH-elimination, and DOX-induced chemotherapy to effectively inhibit tumor growth with minimal side effects in vivo.
