Nanotransferrin-Based Programmable Catalysis Mediates Three-Pronged Induction of Oxidative Stress to Enhance Cancer Immunotherapy

Shuang Bai et al.

Summary: The genetically engineered transferrin-expressing cell membrane nanovesicle encapsulated IR820-dihydroartemisinin nanomedicine (Tf@IR820-DHA) scenario efficiently delivered Fe(III) and IR820-DHA nanoparticles to target tumor locations and induce oxidative stress-associated ICD. This triple stimuli-activated strategy caused high levels of targeted ICD, increasing IFN-gamma-secreting T cell proportions and enhancing a-PD-L1-mediated ICB against primary and distant tumors, representing a promising approach for cancer nanoimmunotherapy.

Tumor-Microenvironment-Activatable Polymer Nano-Immunomodulator for Precision Cancer Photoimmunotherapy

Jing Liu et al.

Summary: This study reports a smart semiconducting polymer nano-immunomodulator that can achieve precise regulation of photodynamic immunotherapy for cancer in the acidic tumor microenvironment. The release of immunogenic factors and TLR7 agonist at the tumor site activates dendritic cells, leading to strong antitumor activities. Additionally, the nano-immunomodulator can directly eradicate tumors and induce immunogenic cancer cell death. This approach enhances systemic antitumor immune responses, resulting in inhibition of tumor growth and metastasis.

Unimolecule-based size-charge switchable nanomedicine for deep cancer sono-immunotherapy

Xiaoxiao Shi et al.

Summary: This study engineered an anti-programmed death ligand 1 peptide inhibitor (APP) to act in the inner core of unimolecular polymers, which could form size-charge switchable nanomicelles in an acidic tumor microenvironment. Under fluorescence/photoacoustic imaging guidance, these nanomicelles could control the exposure of APP and induce reactive oxygen species (ROS) for immune T-cell recruitment and activation in deep solid tumors.

Immunogenic Cell Death Activates the Tumor Immune Microenvironment to Boost the Immunotherapy Efficiency

Zhilin Li et al.

Summary: This article reviews the use of nanostructure-based drug delivery systems (NDDSs) to enhance the effectiveness of tumor immunotherapy. By inducing immunogenic cell death (ICD) and releasing danger-associated molecular patterns (DAMPs) and tumor-associated antigens, in combination with chemotherapy, photodynamic therapy, photothermal therapy, and radiotherapy, the tumor immunosuppressive microenvironment can be improved, resulting in increased sensitivity to immunotherapy and reduced side effects.

Facile Synthesis of a Cubic Porphyrin-Based Covalent Organic Framework for Combined Breast Cancer Therapy

Sainan Liu et al.

Summary: In this research, a cubic porphyrin-based COF named CTP with excellent hydrophilicity was prepared for the first time using a simple method. By linking the porphyrin monomer into the COF, solubility and biocompatibility issues were overcome, and the band gap was narrowed. The resulting CTP showed enhanced sonodynamic effect and photothermal conversion under external irradiation.
