Mitochondria-Targeting Phototheranostics by Aggregation-Induced NIR-II Emission Luminogens: Modulating Intramolecular Motion by Electron Acceptor Engineering for Multi-Modal Synergistic Therapy

Summary: Phototheranostic agents, integrating sensitive in situ fluorescence imaging and effective multi-model synergistic therapy, have emerged as promising tools for cancer theranostics. However, balancing the competitive radiative and nonradiative processes in photon energy transfer remains challenging. In this study, three simple molecules with electron donating-accepting structures were developed. The introduction of an acridinium unit as an electron acceptor led to the formation of TPEDCAc with aggregation-induced second near-infrared emission, high reactive oxygen species generation capability, and excellent photothermal conversion efficiency. The key factors for achieving NIR-II emission and balanced photodynamic/photothermal conversion were found to be large torsional angle and strong electron-withdrawing ability of the acridinium unit. Impressively, TPEDCAc exhibited mitochondria-targeting capability and high performance in in vivo multi-modal cancer theranostics under NIR laser irradiation. This work not only provides a single NIR-II AIE-based multi-modal cancer theranostic system but also inspires new insights into the development of theranostic platforms in the future.

Ultrabright and Highly Polarity-Sensitive NIR-I/NIR-II Fluorophores for the Tracking of Lipid Droplets and Staging of Fatty Liver Disease

Summary: This study developed two bright NIR-I/NIR-II fluorophores with D-pi-A-pi-D structures for tracking fatty liver disease progression in vivo. These fluorophores exhibited ultrahigh quantum yields and ultrasensitive fluorescence response to environment polarity variations. By using these fluorophores as fluorescent nanoprobes, high-resolution imaging of vessels and accurate identification and imaging-guided surgeries of submillimeter scale lymph nodes were successfully achieved. Additionally, the fluorophores showed superior performance in indicating lipid droplets in living cells, enabling precise monitoring and staging of fatty liver disease with a contact-free method utilizing NIR-I/NIR-II fluorescent LDs probe.

Polymerization-Amplified Photoacoustic Signal by Enhancing Near-Infrared Light-Harvesting Capacity and Thermal-to-Acoustic Conversion

Flexible Designing Strategy to Construct Activatable NIR-II Fluorescent Probes with Emission Maxima beyond 1200 nm

Xanthene-Based NIR-II Dyes for In Vivo Dynamic Imaging of Blood Circulation
