Rare SLC13A1 variants associate with intervertebral disc disorder highlighting role of sulfate in disc pathology

Gyda Bjornsdottir et al.

Summary: Back pain is a common and debilitating disorder with largely unknown underlying biology. In this study, a genome-wide association study was conducted using diagnoses assigned in clinical practice, and 41 variants at 33 loci were identified. The most significant association was found with a variant in the CHST3 gene, which is expressed in intervertebral discs. Rare loss-of-function variants in the SLC13A1 gene were also associated with back pain.

Durability of Lubricated Icephobic Coatings under Various Environmental Stresses

Valentina Donadei et al.

Summary: Icephobic coatings have great significance in various industries facing icing problems. However, their durability in real applications is currently limited. Therefore, it is necessary to understand the degradation of coatings under different environmental stresses for further coating development. In this study, lubricated icephobic coatings were fabricated using a flame spray method with hybrid feedstock injection. The performance of the coatings was characterized and investigated under various environmental stresses, including repeated icing/deicing cycles, immersion in corrosive media, and exposure to UV irradiation. The results showed that the coatings exhibited medium-low ice adhesion and slightly more stable icephobic behavior for cottonseed oil-based coatings. Surface roughness slightly increased and wetting performances decreased after cyclic tests, but no chemical changes were observed. The coatings also demonstrated good chemical resistance in selected corrosive media, with better performance for paraffin-based coatings. However, a slight decrease in hydrophobicity was detected due to surface structural changes. The paraffin-based coatings showed better resistance under UV irradiation based on carbonyl index and color change measurements.

Anti-inflammatory and immunomodulatory effects of the extracellular vesicles derived from human umbilical cord mesenchymal stem cells on osteoarthritis via M2 macrophages

Kanglu Li et al.

Summary: This study investigates the immunomodulatory and anti-inflammatory effects of human umbilical cord mesenchymal stem cells-derived extracellular vesicles (hUCMSCs-EVs) on osteoarthritis (OA). The results suggest that hUCMSCs-EVs promote the polarization of M2-type macrophages and the expression of IL-10, inhibit inflammation-associated factors in OA chondrocytes caused by IL-1 beta, and alleviate cartilage degradation during OA progression. The enriched miRNAs in hUCMSCs-EVs and the modulation of the PI3K-Akt signaling pathway may contribute to these therapeutic effects.

Overcoming barriers for intra-articular delivery of disease-modifying osteoarthritis drugs

Jingjing Gao et al.

Summary: Although there has been significant research in intra-articular drug delivery systems and disease-modifying osteoarthritis drugs, there is still a lack of clinically available therapies for osteoarthritis. Multiple barriers hinder the delivery of disease-modifying drugs, but there are promising approaches to overcome these barriers. A patient-centered workflow is proposed to guide the development of intra-articular therapies, with the aim of inspiring innovative solutions for future clinical translation.

Pentraxin 3 regulated by miR-224-5p modulates macrophage reprogramming and exacerbates osteoarthritis associated synovitis by targeting CD32

Jianbin Yin et al.

Summary: An imbalance in M1/M2 macrophage polarization plays a crucial role in the progression of osteoarthritis (OA). Pentraxin 3 (PTX3) is found to be highly expressed in OA M1-polarized macrophages and is involved in the occurrence and development of OA. PTX3 primarily affects OA through macrophages and the paracrine interaction between macrophages and chondrocytes produces a feedback loop that enhances synovitis and cartilage damage. This study identifies a functional pathway important to OA development and suggests that blocking this pathway and PTX3 may prevent and treat OA.
