Summary: The clinical presentation of CADASIL in Greece is similar to Caucasian populations, with a high prevalence of migraine with aura, ischemic cerebrovascular diseases, behavioral-psychiatric symptoms, and cognitive decline. The genetic mutations in Greek CADASIL pedigrees are predominantly found in exon 4, with R169C and R182C mutations being the most common.

Summary: This study described the mutation analysis of the NOTCH3 gene in 24 Portuguese families with suspected CADASIL, revealing different heterozygous variants, including pathogenic cysteine altering variants, cysteine sparing variants, and nonsense variants, mainly located in exons 4, 8, and 11. These genetic findings expand the mutational and clinical spectrum of CADASIL and provide additional evidence for genetic counseling and clinical management.
