Rational Design of Nanotherapeutics Based on the Five Features Principle for Potent Elimination of Cancer Stem Cells

Zhijie Zhang et al.

Summary: Cancer stem cells (CSCs) are a small fraction of tumor cells but play a critical role in tumor initiation, progression, therapy resistance, and recurrence. Traditional CSC-targeting drugs are ineffective against bulk tumor cells and also affect normal stem cells. Nanomaterial drug delivery systems (NDDSs) can passively accumulate in tumor tissues, reducing side effects on normal organs, and can be functionalized for targeted delivery to CSCs. NDDSs have the advantage of synchronized killing of CSCs and bulk cancer cells. This material-based approach for targeting CSCs has gained significant attention recently.

ESE3/EHF, a promising target of rosiglitazone, suppresses pancreatic cancer stemness by downregulating CXCR4

Tianxing Zhou et al.

Summary: EHF plays a key role in decreasing the sensitivity of pancreatic cancer cells to CSCs' niche stimulus by suppressing the transcription of the CXCL12 receptor CXCR4. Rosiglitazone can target PC stem cells and inhibit the crosstalk between CSCs and their niche by upregulating EHF.

Nanomaterial-mediated ablation therapy for cancer stem cells

Yuanwei Pan et al.

Summary: Cancer stem cells (CSCs) are the cause of tumor initiation, metastasis, and relapse. Traditional therapies fail to eliminate CSCs due to their resistance. Nanoparticle-mediated ablation therapies (NMAT) provide new opportunities for eliminating CSCs and have shown promising outcomes.

A nanotherapeutic strategy to overcome chemotherapeutic resistance of cancer stem-like cells

Shiyang Shen et al.

Summary: This study presents a nanoparticle-based strategy for killing CSCs in tumors by inducing CSC differentiation with all-trans retinoic acid release under hypoxic conditions and triggering cell death with camptothecin release in response to increasing reactive oxygen species levels in differentiating CSCs.

Live tumor imaging shows macrophage induction and TMEM-mediated enrichment of cancer stem cells during metastatic dissemination

Ved P. Sharma et al.

Summary: Research has shown that cancer stem cells (CSCs) play an important role in tumor metastasis, with non-stem cancer cells induced to become stem cells through direct contact with macrophages. In human breast cancer, TMEM doorways are not only entry portals for cancer cell intravasation, but also sites for CSC programming.

Stem-like cells drive NF1-associated MPNST functional heterogeneity and tumor progression

Daochun Sun et al.

Summary: NF1-associated MPNSTs originate from benign peripheral nerve neurofibromas and a rare stem-cell-like cell population within MPNSTs plays a crucial role in tumor initiation and relapse. By isolating and characterizing these cells, a cancer stem cell-specific gene expression signature has been identified, with potential therapeutic implications. Targeting cancer stem cells in combination with antimitotic chemotherapy shows promise in inhibiting tumor growth and prolonging survival, with implications for MPNST therapy development.

Small extracellular vesicles in cancer

Komal Abhange et al.

Summary: Extracellular vesicles (EVs) are lipid-bilayer enclosed vesicles released from cells, carrying various molecules and playing crucial roles in tumors. EVs exhibit expression patterns similar to parental cells, aiding in early cancer diagnosis and treatment monitoring. In addition to research and diagnostics, EVs are utilized as drug delivery nanocarriers and therapeutic targets.
