Notch-triggered maladaptation of liver sinusoidal endothelium aggravates nonalcoholic steatohepatitis through endothelial nitric oxide synthase

Summary: This study investigates the role of endothelial Notch signaling in the development of NASH. The results show that activation of Notch signaling in endothelial cells leads to LSEC maladaptation and worsens NASH symptoms, while inhibition of Notch signaling restores LSEC homeostasis and improves NASH phenotypes. Furthermore, the study reveals that Notch activation exacerbates NASH by inhibiting eNOS transcription, and administration of an eNOS activator alleviates hepatic steatosis and lipid accumulation resulting from Notch activation. Finally, the study demonstrates the potential therapeutic effect of Notch inhibitors in treating NASH.
