Tau Post-translational Modifications: Dynamic Transformers of Tau Function, Degradation, and Aggregation

Carolina Alquezar et al.

Summary: Post-translational modifications play a crucial role in regulating tau protein function, including phosphorylation and acetylation. While these modifications may regulate protein function under normal conditions, they can be hijacked in disease states. Limited understanding exists on how these modifications impact tau protein, but studies indicate their critical role in localization, interactions, and other aspects.

Mass-sensitive particle tracking to elucidate the membrane-associated MinDE reaction cycle

Tamara Heermann et al.

Summary: An iSCAT image processing and analysis strategy enables mass-sensitive particle tracking (MSPT) of single unlabeled biomolecules on a supported lipid bilayer, allowing real-time observation of membrane complexes. This approach was applied to study the reaction cycles of MinDE protein self-organization, revealing the potential of MSPT to enhance quantitative understanding of membrane-associated biological systems.

Single molecule mass photometry reveals the dynamic oligomerization of human and plant peroxiredoxins

Michael Liebthal et al.

Summary: Protein oligomerization plays a central role in biological function and regulation, with the challenge of quantifying and measuring dynamic transitions in solution. Mass photometry demonstrates the quantification of affinity and polydispersity of heterogeneous protein complexes in solution, highlighting diverse functionality of oligomerization for 2CPs. The study reveals both common features and species-specific differences in oligomeric transitions, emphasizing the power of mass photometry for characterizing heterogeneous oligomeric protein distributions near native conditions.

Hsp40s play complementary roles in the prevention of tau amyloid formation

Rose Irwin et al.

Summary: This study elucidates the effects of Hsp40 molecular chaperones DNAJA2 and DNAJB1, as well as the small heat shock protein HSPB1, on tau amyloid-fiber formation, demonstrating that they play complementary roles in preventing tau aggregation and interact with distinct sets of tau species. The mechanisms by which these chaperones counteract toxic tau aggregates are further outlined, highlighting the diverse and complementary roles of chaperones from different families/classes in preventing pathological protein aggregation.
