Selective inactivation of Gram-positive bacteria in vitro and in vivo through metabolic labelling

Tao Feng et al.

Summary: Bacterial infections are a serious threat to human health, especially those caused by common Gram-positive bacteria. Photothermal therapy (PTT) has shown potential as a universal antimicrobial technique without inducing drug resistance, but its unspecific killing capability and side effects hinder practical applications. A metabolic engineering strategy using a bioorthogonal photothermal agent was proposed to selectively inactivate Gram-positive bacteria by pre-metabolizing with 3-azido-D-alanine, demonstrating efficient eradication in vitro and in a mouse model. This strategy could lead to the development of precise antibacterial candidates for preclinical applications.

Multifunctional Photoactive Hydrogels for Wound Healing Acceleration

Aziz Maleki et al.

Summary: Light-based photothermal therapy (PTT) and photo-dynamic therapy (PDT) have great potential in wound healing and infection prevention. Multifunctional photoresponsive hydrogels (MPRHs) combining light and hydrogel advantages are increasingly used in biomedicine, particularly in wound repair. The modulation of bacterial microenvironments to inhibit infection through PTT, PDT, and photothermal/photodynamic synergistic therapy has been discussed.

Click and Bio-Orthogonal Reactions with Mesoionic Compounds

Karine Porte et al.

Summary: Click chemistry and bio-orthogonal reactions are primarily based on cycloaddition reactions, especially 1,3-dipolar cycloadditions. Mesoionic compounds have recently emerged as attractive dipole partners due to their ability to be derivatized and have their reactivity tuned towards cycloaddition reactions. Some compounds from the mesoionic family can also undergo click-and-release reactions, showing interesting applications in cells and animals.

Negatively Charged Sulfur Quantum Dots for Treatment of Drug-Resistant Pathogenic Bacterial Infections

Yi Wang et al.

Summary: The study engineered negatively charged sulfur quantum dots (SQDs) as an efficient broad-spectrum antibiotic to kill drug-resistant bacteria in vitro and in vivo. SQDs destroy bacterial membrane systems and affect their metabolism through the intrinsic antibacterial activity of elemental sulfur and catalytic generation of reactive oxygen species. Compared to traditional positively charged materials, SQDs demonstrate better hemocompatibility and lower toxicity.
