p53 in ferroptosis regulation: the new weapon for the old guardian

Summary: A new porous yolk shell nanostructure, PYSNPs, has been reported to stabilize Fe(0) in normal physiological conditions and control its release in the tumor microenvironment for enhanced cancer therapy. These PYSNPs exhibit superior tumor inhibition and can effectively suppress tumor growth in vivo with minimal side effects. Additionally, the disintegration of PYSNPs in the acidic tumor microenvironment can lead to significant changes in MRI signals for contrast-enhanced diagnosis.

p53 Promoted Ferroptosis in Ovarian Cancer Cells Treated with Human Serum Incubated-Superparamagnetic Iron Oxides

Summary: The study demonstrates that a combination of SPIO-Serum and wild-type p53 overexpression can decrease ovarian cancer cell viability by inducing iron-dependent oxidative death and lipid peroxidation. SPIO-Serum treatment disrupts intracellular iron homeostasis, causes mitochondrial structural abnormalities, and involves p53 in ferroptosis induction in ovarian cancer cells.

The toxicity mechanism of different sized iron nanoparticles on human breast cancer (MCF7) cells

Summary: The toxicity mechanism of superparamagnetic iron oxide nanoparticles (SPIONs) was investigated to reduce risks, with higher dosage and suitable sizes of SPIONs enhancing uptake into cells and leading to increased reactive oxygen species (ROS) formation. Smaller SPIONs tended to produce more ROS in mitochondria, damaging them, while larger SPIONs promoted ROS production in plasma to destroy cytomembrane. Different size SPIONs activated distinct cell death pathways, with ferroptosis playing a significant role in mitochondrial and cytomembrane damage. ROS levels correlated with gene expression, suggesting RNA-seq as a new method to assess oxidative stress and toxicity of nanomaterials.
