A DNA Nanostructure-Based Neuroprotectant against Neuronal Apoptosis via Inhibiting Toll-like Receptor 2 Signaling Pathway in Acute Ischemic Stroke

Mi Zhou et al.

Summary: The study demonstrated that tFNA effectively prevented neuronal apoptosis in ischemic stroke by interfering with ischemic cascades in vitro and ameliorating the microenvironment of the ischemic hemisphere in vivo. This pleiotropic nanoneuroprotectant shows promise as a therapeutic strategy for ischemic stroke by attenuating neurological deficits and reducing infarction volume.

Crossing the Blood-Brain Barrier: Advances in Nanoparticle Technology for Drug Delivery in Neuro-Oncology

Andrew M. Hersh et al.

Summary: This article provides a review of the potential of nanoparticles (NPs) in improving drug delivery to brain tumors. The unique physical and chemical properties of NPs and the mechanisms of NP transport across the blood-brain barrier are discussed, along with the major types of NPs investigated for brain tumor treatment. In addition, the use of NPs as imaging contrast agents and their conjugation with imaging probes for visualization and treatment monitoring are explored. Finally, the limitations of NPs for brain tumor treatment are discussed.

Anti-glioblastoma effects of phenolic variants of benzoylphenoxyacetamide (BPA) with high potential for blood brain barrier penetration

Joanna Stalinska et al.

Summary: Glioblastomas are aggressive brain tumors with limited therapeutic options. This study aimed to find new variants of metabolic compounds with anti-glioblastoma activity. Computational analyses were performed to select compounds for further evaluation. Two compounds, HR51 and HR59, demonstrated improved capability of crossing the blood-brain barrier.

Therapeutic hypothermia augments the restorative effects of PKC-β and Nox2 inhibition on an in vitro model of human blood-brain barrier

Rais Reskiawan A. Kadir et al.

Summary: Therapeutic hypothermia enhances the protective effects of PKC-beta or Nox2 inhibition on BBB integrity and function in the setting of hyperglycaemia. Combining PKC-beta or Nox2 inhibition with therapeutic hypothermia is more effective in repairing physical damage, decreasing Nox activity and superoxide anion production, and suppressing actin stress fibre formation compared to using either agent alone. Additionally, the combinatory approach is more effective in maintaining normal cytoskeletal structure.
