3′-Amino modifications enhance the antifungal properties of N4-alkyl-5-methylcytidines for potential biocides

Summary: A new generation of biocides with a broad spectrum of activities against both bacteria and fungi has been developed. By synthesizing novel 3'-modified derivatives of N-4-alkyl-5-methyl-2',3'-dideoxycytidines, compounds were obtained that significantly enhanced antifungal activity, particularly against Aspergillus genus. These compounds showed complete inhibition of the growth of all tested moulds isolated from works of art. In addition, they demonstrated inhibitory effects on bacterial growth, with minimum inhibitory concentrations slightly higher than current therapeutic drugs.

Discovery of novel N4-alkylcytidines as promising antimicrobial agents

Summary: A series of new N-4-cytidine derivatives were synthesized, showing activity against various Gram-positive bacteria and fungi. Among them, N-4-dodecyl-5-methyl-2'-deoxycytidine 4h exhibited high inhibitory activity against Mycobacterium smegmatis, Staphylococcus aureus, and filamentous fungi that can damage artworks.

Engineering the Steroid Hydroxylating System from Cochliobolus lunatus in Mycolicibacterium smegmatis

Summary: A synthetic bacterial operon containing the cytochrome P450 CYP103168 and the reductase CPR64795 of the fungus Cochliobolus lunatus has been engineered into a shuttle plasmid, which was then used to transform Mycolicibacterium smegmatis mutants for efficient expression of the hydroxylating CYP system and production of hydroxylated steroids. This new approach allows for direct synthesis of hydroxylated steroidal products from cholesterol and phytosterols in a single fermentation step, offering a potential avenue for industrial-scale production of these compounds.
