The Potential of ANGPTL8 Antagonism to Simultaneously Reduce Triglyceride and Increase HDL-Cholesterol Plasma Levels

Summary: Elevated triglyceride and reduced high-density lipoprotein-cholesterol plasma levels are risk factors for atherosclerosis and cardiovascular disease. By inhibiting lipoprotein lipase and endothelial lipase, Angiopoietin-like 3 (ANGPTL3) plays a role in regulating plasma triglyceride and HDL-C levels. Angiopoietin-like 3 inhibition of LPL requires complex formation with Angiopoietin-like 8, which is not required for its inhibition of EL. By disrupting or precluding the ANGPTL3-8 complex formation, ANGPTL8 antagonism can increase LPL activity while decreasing EL activity, leading to reduced plasma triglyceride and increased HDL-C levels.
