Dynamic closed states of a ligand-gated ion channel captured by cryo-EM and simulations

Summary: This study presents cryo-EM structures of the proton-activated Gloeobacter violaceus ligand-gated ion channel (GLIC) under three pH conditions, revealing improved resolution and rearrangements at the subunit/domain interface with decreased pH. Molecular dynamics simulations indicate flexibility in the closed-channel extracellular domains and support electrostatic remodeling during proton-induced gating. Exploration of secondary cryoEM classes shows a low-pH population with an expanded pore, defining distinct protonation and activation steps in GLIC's pH-stimulated conformational cycling.
