Cardiac glycosides cause cytotoxicity in human macrophages and ameliorate white adipose tissue homeostasis

Antoni Olona et al.

Summary: The study shows that cardiac glycosides such as ouabain can induce cytotoxicity in macrophages and have potential effects on human tissues. Ouabain triggers pyroptosis and apoptosis, with a stronger effect on monocyte-derived macrophages and influenced by K+ flux. Furthermore, treatment with ouabain in adipose tissue from obese patients results in improved insulin sensitivity and reduction of macrophages.

Adoptive transfer of immunomodulatory M2 macrophages suppresses experimental autoimmune encephalomyelitis in C57BL/6 mice via blockading NF-κB pathway

F. Chu et al.

Summary: Macrophages play crucial roles in the pathogenesis of EAE, with M2 macrophages having anti-inflammatory effects. Treatment with M2 macrophages can successfully block the NF-kappa B pathway and alleviate symptoms in EAE patients. Application of NF-kappa B blockers is beneficial in preventing and treating EAE.

Therapeutic potential of TNFa inhibitors in chronic inflammatory disorders: Past and future

Hongtao Zhang et al.

Summary: The availability of biological TNFa inhibitors has significantly advanced the treatment of immune diseases such as rheumatoid arthritis in the past 20 years. However, there are still unmet needs including side effects, tolerance issues, and the development of new TNFa inhibitors focusing on TNFR1-specific inhibitors.

Risk for infections with glucocorticoids and DMARDs in patients with rheumatoid arthritis

Thomas R. Riley et al.

Summary: Immunomodulatory therapy for rheumatoid arthritis carries infectious complications risk. Different therapeutic options have varying risk levels, with TNF inhibitors showing an increase in serious infections risk. Other biological agents and JAK inhibitors have similar risk levels, with JAK inhibitors also associated with higher risk of herpes zoster.
