Improved synthesis of an ergothioneine PET radioligand for imaging oxidative stress in Alzheimer's disease

William J. Behof et al.

Summary: This study presents an efficient method for the preparation of a [C-11]-labeled ERGO PET radioligand, which is able to detect threshold levels of oxidative stress in a preclinical animal model of Alzheimer's disease.

Ex vivo MRI and histopathology detect novel iron-rich cortical inflammation in frontotemporal lobar degeneration with tau versus TDP-43 pathology

M. Dylan Tisdall et al.

Summary: Frontotemporal lobar degeneration (FTLD) encompasses a range of neurodegenerative diseases characterized by proteinopathies, including Tau and TDP-43. Clinical differentiation between these subtypes is challenging, but integrated MRI and histopathology studies may help distinguish FTLD-Tau and FTLD-TDP.

Effect of Ergothioneine on 7-Ketocholesterol-Induced Endothelial Injury

Sally Shuxian Koh et al.

Summary: Ergothioneine (ET) has a protective effect on human brain endothelial cells, reducing apoptosis, necrosis, and inflammation induced by 7-ketocholesterol (7KC). ET exerts its anti-inflammatory effects by reducing the expression of pro-inflammatory cytokines IL-1beta, IL-6, IL-8, TNF-alpha, and cyclooxygenase-2 (COX2).

A novel antioxidant ergothioneine PET radioligand for in vivo imaging applications

William J. Behof et al.

Summary: This study successfully demonstrated the biodistribution of ERGO in vivo using a [C-11]ERGO PET radioligand, showing significant accumulation in the brain, eyes, and small intestine of the mice. The radioligand also remained strong in the brain 30 minutes post-injection, marking the first preclinical demonstration of ERGO's biodistribution in a highly sensitive and non-invasive manner.
