Global coagulation assays in hypercoagulable states

Hui Yin Lim et al.

Summary: Thrombosis is a significant global cause of disease and death, and accurately predicting the risk of thrombotic and cardiovascular complications remains a challenge in modern medicine. Conventional coagulation tests lack sufficient information, driving the exploration of global coagulation assays. However, their role in thrombotic disorders requires further investigation.

Endothelial function in cardiovascular medicine: a consensus paper of the European Society of Cardioloy working Groups on Artherosclerosis and Vascular Biology, Aorta and Perirpheral Vascular Diseases, Coronary Pathophysiology and Microcirculation, and Thrombosis

Yvonne Alexander et al.

Summary: Endothelial cells play a crucial role in cardiovascular health, and dysfunction can be caused by cardiovascular risk factors but restored once stimuli are removed. There are various methods to measure endothelial function, with each having its pros and cons. It is recommended to adopt a consensus methodology like flow-mediated dilatation and establish reference values for different populations. Future studies should explore the integration of endothelial function measurements with patient-specific epigenetic data and other biomarkers.

Assessing Plasmin Generation in Health and Disease

Adam Miszta et al.

Summary: Fibrinolysis is a crucial process in wound healing, with plasmin playing a central role in dissolving clots. Recent advancements have enabled the quantification of plasmin generation (PG) and inhibition kinetics during fibrinolysis. Understanding plasmin kinetics aids in identifying fibrinolytic dysfunction and investigating the link between abnormal fibrin dissolution and disease pathogenesis.

Global coagulation assays in healthy controls: are there compensatory mechanisms within the coagulation system?

Hui Yin Lim et al.

Summary: This study evaluated the hemostatic profile of healthy controls using GCAs and found that females exhibited more hypercoagulable parameters in TEG and CAT, while individuals over 50 years old showed more hypercoagulable TEG parameters. Interestingly, individuals with reduced thrombin generation were associated with increased cardiovascular risk factors, possibly due to increased TFPI levels.

Genetics of Cardiovascular Disease: How Far Are We from Personalized CVD Risk Prediction and Management?

Michal Vrablik et al.

Summary: Despite advances in molecular genetics and new tools for identifying new genes and variants related to cardiovascular disease (CVD), there is still some ambiguity in using genetic risk scores for predicting CVD risk. Future research should focus on regulatory molecules and epigenetic changes to further understand genetic factors contributing to CVD.

SCORE2 risk prediction algorithms: new models to estimate 10-year risk of cardiovascular disease in Europe

Steven Hageman et al.

Summary: The study aimed to develop, validate, and demonstrate an updated prediction model (SCORE2) to estimate 10-year cardiovascular disease risk in individuals without previous history of CVD or diabetes in Europe. The new algorithm, SCORE2, enhances the identification of individuals at higher risk of developing CVD across Europe by predicting first-onset CVD risk in European populations.

Effects of Hyperglycemia and Diabetes Mellitus on Coagulation and Hemostasis

Summary: Patients with diabetes are prone to a prothrombotic state due to metabolic disorders, which can be managed with appropriate anticoagulation therapies and metabolic optimizations. Current guidelines lack recommendations on the optimal antithrombotic treatment for these patients.

Global coagulation assays in patients with diabetes mellitus

Hui Yin Lim et al.

Summary: The study found that patients with diabetes have more hypercoagulable profiles on global coagulation assays, especially those with type 2 diabetes and microvascular complications. The results suggest that global coagulation assays may be useful in predicting long-term outcomes for patients.

Clonal hematopoiesis of indeterminate potential (CHIP) and cardiovascular diseases-an updated systematic review

Nagendra Boopathy Senguttuvan et al.

Summary: The presence of clonal hematopoiesis of indeterminate potential (CHIP) mutations is associated with an increased risk of cardiovascular diseases, with DNMT3A, TET2, ASXL1, TP53, JAK2, and SF3B being commonly implicated genes. The analysis of commonly mutated CHIP genes using bioinformatics tools revealed significant interactions in molecular functions and biological processes.
