SOX17 integrates HOXA and arterial programs in hemogenic endothelium to drive definitive lympho-myeloid hematopoiesis

Ho Sun Jung et al.

Summary: The research findings suggest that SOX17 is a key regulator in linking arterial and HOXA programs in hemogenic endothelium (HE) in humans, playing a crucial role in the specification of HE with robust lympho-myeloid potential. This understanding may contribute to better programming of hematopoietic stem cell fate from human pluripotent stem cells.

Sequence logic at enhancers governs a dual mechanism of endodermal organ fate induction by FOXA pioneer factors

Ryan J. Geusz et al.

Summary: FOXA plays a dual role in endodermal organ development: facilitating signal-dependent lineage initiation via enhancer priming, and enforcing organ cell type-specific gene expression via indirect recruitment by lineage-specific TFs. Only a small subset of organ-specific enhancers are bound and primed by FOXA prior to lineage induction, whereas the majority engage FOXA upon lineage induction without undergoing chromatin priming.
