Summary: In this study, we investigated the clinical-epidemiological, microbiological, and molecular aspects of feline sporotrichosis in Espirito Santo, Brazil, and evaluated the reliability of cytopathological examination in diagnosis. The results showed that cytopathological examination demonstrated high agreement and accuracy, and there was an overwhelming occurrence of Sporothrix brasiliensis in the region.

An overview of using fungal DNA for the diagnosis of invasive mycoses

Summary: Fungal PCR has been standardized and is ready for widespread use in the screening and diagnosis of invasive fungal disease (IFD). PCR testing is recommended for infections caused by Aspergillus, Candida, and Pneumocystis jirovecii, and combination with serological testing can enhance the diagnosis. Molecular diagnostics represent a non-classical approach for other IFD and have improved confidence in testing through continued validation and standardization. Molecular testing can also diagnose antifungal resistance, and next-generation sequencing has the potential to improve our understanding of fungal phylogeny, epidemiology, and resistance mechanisms.

Summary: Sporotrichosis is a chronic fungal disease that affects humans and animals. This study aimed to evaluate and validate a nested PCR technique for detecting Sporothrix sp. DNA from formalin-fixed and paraffin-embedded tissues (FFPE). The results showed that the chemical extraction protocol with 5μm thick paraffin sections was the most effective in extracting DNA from Sporothrix sp. The nested PCR technique exhibited high sensitivity and specificity for detecting Sporothrix sp. in FFPE samples.

Treatment can be challenging due to resistance, but alternative drugs hold promise. This review explores these trends to mitigate the emerging and re-emerging disease.

Biofilm formation on cat claws by Sporothrix species: An ex vivo model

Summary: This study evaluated the ability of Sporothrix species to form biofilms on the surface of cat claws using an ex vivo model. Results showed that all species were able to form biofilms on cat claws, with metabolic activity similar to in vitro biofilms. This is the first report of an ex vivo biofilm model involving cat claws, which could increase the viable period of the fungus and the number of potentially infected animals and people.
