Cancer statistics for African American/Black People 2022

Angela N. Giaquinto et al.

Summary: African American/Black individuals bear a disproportionate cancer burden, with high mortality and low survival rates compared to other racial/ethnic groups. Data from the American Cancer Society shows that in 2018, there were significant disparities in cancer incidence, mortality, survival, screening, and risk factors among Black people in the United States. The projected numbers for 2022 indicate a high number of new cancer cases and deaths among Black individuals. Black men have higher incidence and mortality rates for certain cancers, such as myeloma, stomach cancer, and prostate cancer. The disparity in overall cancer mortality between Black and White men is narrowing, but the decline in prostate cancer mortality among Black men has slowed. Black women have a lower incidence rate but higher mortality rates for certain cancers, such as endometrial and breast cancer. Breast cancer has become the leading cause of cancer death among Black women.

IGF2BP2 promotes gastric cancer progression by regulating the IGF1R-RhoA-ROCK signaling pathway

Dong Liu et al.

Summary: Our study aimed to investigate the molecular mechanism of IGF2BP2 in gastric cancer (GC). IGF2BP2 was found to be significantly elevated in both GC tissues and cells. Silencing of IGF2BP2 suppressed inflammation, proliferation, migration and invasion, and promoted cell apoptosis. IGF2BP2, as a m6A reader, increased the expression of IGF1R by identifying m6A methylation modification sites in IGF1R mRNA, thus activating RhoA-ROCK pathway. The anti-carcinogenic impacts of IGFBP2 silence were restrained by IGF1R overexpression, which was eliminated by the inactivation of RhoA-ROCK. Our findings highlight the oncogenic role of IGF2BP2 in gastric carcinogenesis and its potential as a prognostic biomarker.

METTL1 promotes hepatocarcinogenesis via m7G tRNA modification-dependent translation control

Zhihang Chen et al.

Summary: The N-7-methylguanosine (m(7)G) modification is a common transfer RNA (tRNA) modification in humans. Elevated levels of METTL1 and WDR4 in hepatocellular carcinoma (HCC) are associated with advanced tumor stages and poor patient survival. Mechanistically, METTL1-mediated tRNA m(7)G modification promotes the translation of target mRNAs with higher frequencies of m(7)G-related codons, providing new insights into the role of misregulated tRNA modifications in liver cancer.

N7-Methylguanosine tRNA modification enhances oncogenic mRNA translation and promotes intrahepatic cholangiocarcinoma progression

Summary: Intrahepatic cholangiocarcinoma (ICC) shows upregulation of N-7-methylguanosine (m(7)G) tRNA modification and its methyltransferase complex components, METTL1 and WDR4, which are associated with poor prognosis. METTL/WDR4 play a critical role in promoting ICC cell survival and progression by selectively regulating the translation of oncogenic transcripts through m(7)G-tRNA-decoded codon-frequency-dependent mechanisms. Studies with overexpression and knockout mouse models demonstrate the crucial oncogenic function of Mettl1-mediated m(7)G tRNA modification in promoting ICC tumorigenesis and progression in vivo.
