Identification of key microRNAs and the underlying molecular mechanism in spinal cord ischemia-reperfusion injury in rats

Summary: Spinal cord ischemia-reperfusion injury (SCII) is a pathological process that can lead to severe complications such as paraplegia and paralysis. Aberrant miRNA expression is involved in the development of SCII, and different experimental conditions can lead to varied miRNA expression results. This study identified 19 key differently expressed miRNAs (DEmiRNAs) in SCII, with 9 upregulated and 10 downregulated DEmiRNAs. Functional and transcription factor analyses revealed that the upregulated DEmiRNAs mainly targeted the cGMP-PKG and cAMP signaling pathways, while the downregulated DEmiRNAs targeted the Chemokine and MAPK signaling pathways. Among these DEmiRNAs, miR-3568 stood out as a potential intervention target for SCII, as its inhibition preserved hind limb function by reducing apoptosis and regulating key genes such as GATA6, GATA4, and RBPJ.
