Functional CDKN2A assay identifies frequent deleterious alleles misclassified as variants of uncertain significance

Summary: Pathogenic germline CDKN2A variants were found to be associated with an increased risk of pancreatic ductal adenocarcinoma (PDAC). The study discovered that over 40% of CDKN2A variants of uncertain significance (VUSs) identified in PDAC patients were functionally deleterious and likely pathogenic. Reclassifying these functionally deleterious VUSs increased the reported prevalence of pathogenic/likely pathogenic CDKN2A in PDAC patients to up to 4.1%, depending on family history. These findings suggest that CDKN2A VUSs may play a significant, underappreciated role in the risk of pancreatic cancer.
