Transition to invasive breast cancer is associated with progressive changes in the structure and composition of tumor stroma

Tyler Risom et al.

Summary: This study used MIBI-TOF and antibody staining methods to analyze the changes in the tumor microenvironment (TME) between DCIS and IBC. Four coordinated transitions between TME states were identified. The study also found that myoepithelial disruption was more advanced in DCIS patients who did not develop IBC, suggesting a potential protective effect against recurrence.

The breast pre-cancer atlas illustrates the molecular and microenvironmental diversity of ductal carcinoma in situ

Daniela Nachmanson et al.

Summary: The progression of Breast Ductal Carcinoma In Situ (DCIS) is influenced by microenvironmental and molecular factors, which are not well understood. This study analyzed multiple microdissected regions from 39 cases and characterized the mutational, transcriptional, histological, and microenvironmental landscape. Genetic divergence increased with physical distance and phenotypic and subtype heterogeneity was associated with underlying genetic heterogeneity. The study also identified an immune escape state with histological and molecular features located preferentially at DCIS regions.

The harsh microenvironment in early breast cancer selects for a Warburg phenotype

Mehdi Damaghi et al.

Summary: The harsh microenvironment of ductal carcinoma in situ (DCIS) selects for a Warburg Effect (WE) phenotype through constitutive transcriptional reprogramming, with KLF4 potentially being an inducer of the WE phenotype. In vitro results were used to simulate DCIS phenotypic evolution using a mathematical model.

SCDC: bulk gene expression deconvolution by multiple single-cell RNA sequencing references

Meichen Dong et al.

Summary: SCDC is a deconvolution method for bulk RNA-seq data that improves the accuracy of cell-type decomposition. It leverages cell-type specific gene expression profiles from multiple scRNA-seq reference datasets and integrates deconvolution results from different experiments and laboratories. The study demonstrates that SCDC outperforms existing methods in both settings.
