Quantitative Label-Free Proteomic Analysis of Milk Fat Globule Membrane in Donkey and Human Milk

Summary: Previous studies have suggested that donkey milk shares similar compositions with human milk and could be a hypoallergenic alternative for infants with cow's milk allergy. This study used label-free proteomics to compare differentially expressed milk fat globule membrane (MFGM) proteins in donkey milk and human milk, identifying differences in immune system regulation, membrane invagination, and lymphocyte activation. The findings contribute to understanding the biological functions and potentially valuable information for nutritional quality assessment.

Summary: Protein expression profiles were analyzed in Beijing-You chickens at different chronological ages, revealing that the profiles at 120 days were similar to 150 days but distinct from 90 days, suggesting 120 days as a crucial age point. Differentially expressed proteins were mainly involved in key pathways such as glycolysis/gluconeogenesis and focal adhesion. Some proteins were validated using parallel reaction monitoring.

Summary: Through LC-MS/MS and TMT labeling analysis, 123 differentially abundant proteins related to the differentiation process of goat intramuscular adipocytes were identified. Further analyses revealed that the cholesterol metabolism pathway, glucagon signaling pathway, and glycolysis/gluconeogenesis pathway may play important roles in goat intramuscular adipocytes differentiation. Identified proteins such as SRSF10, CSRP3, APOH, PPP3R1, CRTC2, FOS, SERPINE1, and AIF1L could be potential candidates for elucidating the molecular mechanism of intramuscular fat differentiation in goats.
