Identification of 7,8-dihydroxy-3-phenylcoumarin as a reversible monoamine oxidase enzyme inhibitor

Summary: In this study, we evaluated the potential of 3-arylcoumarin derivatives as human monoamine oxidase-A and -B inhibitors, finding that compound 3j was the most effective in inhibiting both MAO-A and MAO-B enzymes. Compound 3j showed nonselectivity, reversible inhibition, and neuroprotection against H2O2-treated human neuroblastoma cells. Molecular modeling suggested that compound 3j's inhibitory activity may be due to its orientation within the enzyme active site, making it a promising scaffold for the development of MAO inhibitors.

Gamma-decanolactone: Preliminary evaluation as potential antiparkinsonian drug

Summary: The study found that gamma-decanolactone can selectively and reversibly inhibit MAO-B in vitro and is safe at higher concentrations, with potential as an antiparkinsonian drug. In vivo experiments showed that gamma-decanolactone partially reversed ataxia in reserpinized mice, and concomitant treatment with Levodopa plus Benserazide enhanced this effect.

Summary: SHQA inhibits the activity of COX-2, reduces the secretion of inflammatory mediators, and suppresses NF-κB activation by preventing I κB-α degradation and inhibiting Akt phosphorylation. Additionally, SHQA induces heme oxygenase-1 expression via Nrf2 activation. These findings suggest that SHQA could be a potential therapeutic agent for various inflammatory diseases.
