Downregulation of ROCK2 attenuates alcohol-induced inflammation and oxidative stress in astrocytes

Xinguo Li et al.

Summary: This study demonstrated the role of ROCK2 in alcohol-induced injury to astrocytes. Downregulation of ROCK2 alleviated astrocytic injury by reducing inflammation and oxidative stress. Moreover, downregulation of ROCK2 inhibited the activation of NF-kappa B signaling pathway and NLRP3 inflammasome, suggesting that ROCK2 could be a potential therapeutic target for alcohol-induced astrocytic injury.

Concurrent suppression of Aβ aggregation and NLRP3 inflammasome activation for treating Alzheimer's disease

Tao Yang et al.

Summary: Alzheimer's disease (AD) is a neurodegenerative illness characterized by memory loss, cognitive disorders, and impaired behavior. A beta plaques and NLRP3 inflammasome activation play important roles in its pathogenesis. A bifunctional molecule, BPBA, has been developed to target A beta and inhibit NLRP3 inflammasome, effectively dissolving A beta aggregates and reducing neuroinflammation.

Methylsulfonylmethane protects against ethanol-induced brain injury in mice through the inhibition of oxidative stress, proinflammatory mediators and apoptotic cell death

Keyvan Amirshahrokhi et al.

Summary: Excessive ethanol consumption can cause brain injury, but this study found that methylsulfonylmethane (MSM) has a protective effect against ethanol-induced brain injury. MSM reduces oxidative stress and inflammation, increases antioxidant levels, and decreases apoptosis-related proteins. These findings suggest that MSM may be a potential approach to prevent brain damage caused by high levels of alcohol.

SOCE-mediated NFAT1-NOX2-NLRP1 inflammasome involves in lipopolysaccharide-induced neuronal damage and A beta generation

Zhenghao Sun et al.

Summary: The presence of lipopolysaccharide (LPS) is associated with higher levels of LPS in the blood and brains of Alzheimer's disease (AD) patients, which is linked to neuronal damage and the generation of beta-amyloid (A beta). This study investigated the mechanism by which LPS causes neuronal damage and found that oxidative stress, neuroinflammation, and Ca2+ overload play important roles in AD. The activation of SOCE-mediated NFAT1-NOX2-NLRP1 inflammasome is involved in LPS-induced neuronal damage and A beta generation. Treatment with inhibitors and scavengers reversed these LPS-induced changes and improved neuronal damage and A beta deposition.

Neuronal NLRP3 is a parkin substrate that drives neurodegeneration in Parkinson's disease

Nikhil Panicker et al.

Summary: Parkinson's disease is caused by alpha-synuclein accumulation and death of dopamine neurons in the substantia nigra pars compacta, with NLRP3 inflammasome hyperactivation playing a role in the disease. Loss of parkin activity leads to spontaneous assembly of NLRP3 inflammasome in dopamine neurons, which is further exacerbated by mitochondrial-derived reactive oxygen species. Inhibition of neuronal NLRP3 inflammasome assembly can prevent degeneration of dopamine neurons in PD models.
