Apathy is associated with white matter network disruption and specific cognitive deficits in Parkinson's disease

Ming-Ching Wen et al.

Summary: This study investigated the impact of apathy on cognitive functions and the underlying white matter network alterations in Parkinson's disease (PD). The results showed that apathetic PD patients had impaired neural integration and reduced structural connectivity. Apathy, depression, and motor severity showed distinct impacts on different cognitive functions, with apathy being the most influential determinant of cognition in PD.

The 3-Item Apathy Subscale Within the GDS-15 Is Not Supported in De Novo Parkinson's Disease Patients: Analysis of the PPMI Cohort

Sarah M. Szymkowicz et al.

Summary: This study aimed to determine the applicability of the 3-item Withdrawal-Apathy-Lack of Vigor (WAV) subscale in newly diagnosed Parkinson's disease patients, finding it not supported in this sample. While the internal consistency of GDS-15 was acceptable, data reduction techniques revealed components different from the reported subscale. The study concluded that the existing factor structures of GDS-15 were not suitable for this sample.

Physical Activity Improves Anxiety and Apathy in Early Parkinson's Disease: A Longitudinal Follow-Up Study

Samuel Yong-Ern Ng et al.

Summary: The study found that higher baseline physical activity in early PD patients was associated with younger age, lower MDS-UPDRS motor scores, smaller levodopa equivalent daily dose, better attention and memory scores, and better quality of life. Additionally, it was revealed that higher baseline physical activity was associated with decreased anxiety and apathy scores at 1-year follow-up.

Serotonergic and Dopaminergic Lesions Underlying Parkinsonian Neuropsychiatric Signs

Audrey Maillet et al.

Summary: The severity of dopaminergic and serotonergic dysfunction in Parkinson's disease worsens with disease duration, spreading from midbrain/subcortical to cortical regions. Early onset apathy is associated with more severe dopaminergic and serotonergic disruption, similar to intermediate disease duration patients. The severity of neuropsychiatric symptoms appears primarily related to serotonergic alteration within corticostriatal limbic areas.

Longitudinal evolution of non-motor symptoms in early Parkinson's disease: a 3-year prospective cohort study

Ruwei Ou et al.

Summary: The study of global non-motor symptoms (NMS) in Chinese patients with Parkinson's disease (PD) showed an increase in the majority of NMS domains except for mood/apathy and sexual dysfunctions. The evolution of overall NMS in early PD was found to be mild and multidimensional, with a high NMS burden predicting faster motor progression of PD. This longitudinal study provides valuable insights into the progression of NMS in PD.

Jared T. Hinkle et al.

Summary: This study found that neuropsychiatric symptoms such as apathy and anxiety at the onset of Parkinson's disease may predict the severity of motor complications associated with dopamine replacement therapy, highlighting the potential role of these symptoms as biomarkers for vulnerability to early and disabling motor complications.

Neuropsychiatric symptoms and striatal monoamine availability in early Parkinson's disease without dementia

Yoon-Sang Oh et al.

Summary: This study found that patients with early Parkinson's disease who exhibited neuropsychiatric symptoms had significantly lower monoamine availability in the thalamus compared to those without neuropsychiatric symptoms. This suggests that decreased monoamine transporter availability in the thalamus may serve as an imaging biomarker for neuropsychiatric symptoms in PD patients.
