Sarecycline treatment for acne vulgaris: Rationale for weight-based dosing and limited impact of food intake on clinical efficacy

Ayman Grada et al.

Summary: The study investigated the impact of food intake on the absorption and clinical efficacy of sarecycline, a new oral narrow-spectrum third-generation tetracycline-class antibiotic approved for acne vulgaris treatment. Co-administration with high-fat food was found to decrease exposure by 21.7%, but had limited impact on clinical efficacy, allowing for weight-based dosing and flexibility in administration.

Sarecycline Demonstrated Reduced Activity Compared to Minocycline against Microbial Species Representing Human Gastrointestinal Microbiota

Mahmoud A. Ghannoum et al.

Summary: This study evaluated the impact of sarecycline on gut microbiota and found that it had reduced antimicrobial activity compared to minocycline against a wide range of microorganisms. This suggests that sarecycline is less disruptive to gut microbiota.

Sarecycline Demonstrates Clinical Effectiveness against Staphylococcal Infections and Inflammatory Dermatoses: Evidence for Improving Antibiotic Stewardship in Dermatology

Ayman Grada et al.

Summary: We evaluated the clinical effectiveness of oral sarecycline against cutaneous staphylococcal infections, eyelid stye, and mucous membrane pemphigoid and found that sarecycline is a viable option for clinicians to practice improved antibiotic stewardship. Our findings demonstrate the superiority of sarecycline over broad-spectrum tetracyclines in treating staphylococcal skin infections and inflammatory skin diseases.

Antibiotic resistance in acne: changes, consequences and concerns

A. S. Karadag et al.

Summary: Antibiotic resistance in acne has been a major concern in dermatology since the 1970s, with significant variations in resistance rates and types of antimicrobials globally. While resistance to certain antibiotics remains a problem worldwide, resistance to others has remained low in the past decade.

Oral Antibiotics for Acne

Dillon J. Patel et al.

Summary: Oral antibiotics are crucial for treating inflammatory acne, but there are concerns about antibiotic resistance and phobia. Dermatologists need to carefully consider dosage, duration, and frequency when prescribing oral antibiotics, and avoid using them as monotherapy. Factors such as concomitant medications, allergies, and disease severity, as well as costs and patient demographics, can influence prescribing plans and variability.

The microbiome and human cancer

Gregory D. Sepich-Poore et al.

Summary: Microbes play significant roles in cancer, and bacteria, viruses, and fungi are believed to be key actors in cancer immunotherapy. Although the number of microbes known to directly cause carcinogenesis is small, further research is needed to understand the causal relationship between microbes and cancer.

Antibacterial Mechanisms and Efficacy of Sarecycline in Animal Models of Infection and Inflammation

Christopher G. Bunick et al.

Summary: Sarecycline, a narrow-spectrum third-generation tetracycline-class antibiotic approved by the FDA for treating moderate-to-severe acne, exhibits activity against Gram-positive bacteria, particularly Staphylococcus aureus, with reduced efficacy against Gram-negative bacteria like Escherichia coli. Its unique C7 moiety interacts with bacterial ribosomes to block translation and prevent resistance, showing promising results in in vivo studies of infection and inflammation.
