Spatially Resolved Transcriptomic Analysis of Acute Kidney Injury in a Female Murine Model

Eryn E. Dixon et al.

Summary: This study utilized a new spatial transcriptomic sequencing platform to analyze the topography of gene expression and revealed intercellular communication networks and regional gene expression patterns during kidney injury and repair. The results showed that injured proximal tubule cells continued to interact with macrophages and lymphocytes even 6 weeks after injury, potentially reflecting the transition from acute kidney injury to chronic kidney disease.

The orchestrated cellular and molecular responses of the kidney to endotoxin define a precise sepsis timeline

Danielle Janosevic et al.

Summary: Sepsis is a dynamic and life-threatening condition that progresses at variable rates. In a murine endotoxemia model, single-cell RNA sequencing revealed temporally organized involvement of different cell populations. Endothelial and stromal cells were the initial responders, while later epithelial cells played a key role in regulating immune-related pathways.

Integrins and extracellular matrix proteins modulate adipocyte thermogenic capacity

Maria A. Gonzalez Porras et al.

Summary: This study revealed that mice with a null mutation in the LAMA4 gene exhibit resistance to obesity and increased expression of thermogenic fat markers in WAT. The levels of ITA7 and LAMA4 decreased during adipose-derived stem cell differentiation into beige fat, with ITA7 playing a crucial role in beige adipose formation.

Phenotypic diversity and metabolic specialization of renal endothelial cells

Sebastien J. Dumas et al.

Summary: Renal endothelial cells exhibit phenotypic and molecular heterogeneity in response to different microenvironments, supporting kidney functions and potentially serving as targets for therapeutic strategies. Understanding the diversity and specialization of kidney endothelial cells could offer new approaches for treating kidney diseases and promoting kidney regeneration.

Single cell transcriptional and chromatin accessibility profiling redefine cellular heterogeneity in the adult human kidney

Yoshiharu Muto et al.

Summary: The integration of single cell transcriptome and chromatin accessibility datasets provides deeper insights into cell heterogeneity, demonstrating the potential of single nucleus ATAC-seq in the identification of cell identity and functional heterogeneity. Differentially accessible chromatin regions in promoters associated with differentially expressed genes suggest the involvement of transcription factors like NF-kappa B in driving transitions between cell subpopulations within the kidney. This multi-omics approach improves the detection of unique cell states and redefines cellular heterogeneity in specific regions of the kidney.

Single cell regulatory landscape of the mouse kidney highlights cellular differentiation programs and disease targets

Zhen Miao et al.

Summary: The study on the epigenetic program in different cell types of the kidney is crucial for understanding cell-type heterogeneity during tissue homeostasis and injury response. By analyzing open chromatin and gene expression in mouse kidneys, key cell type-specific transcription factors and major gene-regulatory circuits for kidney cells are revealed. Additionally, single nucleotide variants associated with human kidney disease implicate specific cell types, developmental stages, genes, and regulatory mechanisms.

A coordinated progression of progenitor cell states initiates urinary tract development

Oraly Sanchez-Ferras et al.

Summary: The development of the kidney and upper urinary tract relies on interactions between the ureteric bud and mesenchyme. The coordinated organization of four major progenitor cell populations during nephric duct morphogenesis is regulated by transcription factors Tfap2a/b and Gata3. This understanding can potentially guide the differentiation of functional kidney tissue.

Integration of spatial and single-cell transcriptomics localizes epithelial cell-immune cross-talk in kidney injury

Ricardo Melo Ferreira et al.

Summary: This study characterized the transcriptomic signature of cell types within the kidney and found that the spatial distribution of acute kidney injury (AKI) affects cells heterogeneously. By mapping different cell types to human nephrectomy, the researchers were able to predict cell-type spots that corresponded with histopathology. Furthermore, the study identified localized regions of reduced overall expression associated with injury pathways in murine AKI models.
