Tetrandrine alleviates silicosis by inhibiting canonical and non-canonical NLRP3 inflammasome activation in lung macrophages

Mei-yue Song et al.

Summary: This study established silicosis mouse models to investigate the effectiveness of tetrandrine in early and late therapeutic administration, revealing its novel mechanism of attenuating silicosis by inhibiting the NLRP3 inflammasome pathways in lung macrophages. The findings not only demonstrated promising results against silicosis-associated inflammation and fibrosis, but also laid the groundwork for understanding the molecular targets of tetrandrine, potentially leading to a globally accepted therapeutic strategy for silicosis.

Molecular mechanisms and therapeutic implications of tetrandrine and cepharanthine in T cell acute lymphoblastic leukemia and autoimmune diseases

Wencheng Xu et al.

Summary: Inappropriately activated T cells can lead to autoimmune diseases and T-ALL. Bisbenzylisoquinoline alkaloids tetrandrine and cepharanthine have shown potential in treating some autoimmune diseases and cancers, but further studies on their mechanisms of action and combined applications with other drugs are needed. These alkaloids influence the growth of activated T cells through various signaling pathways, and their P-glycoprotein inhibitory effect may be beneficial for refractory T cell-related diseases. Combined with glucocorticoids or anti-leukemia drugs, tetrandrine and cepharanthine offer a new therapeutic option for patients with drug-resistant conditions.

A Novel Inflammatory Dendritic Cell That Is Abundant and Contiguous to T Cells in the Kidneys of Patients With Lupus Nephritis

Samir V. Parikh et al.

Summary: The study identified a novel inflammatory dendritic cell (infDC) population highly expressed in the kidneys of patients with proliferative LN. Further investigation revealed that these infDCs interact with T cells in the kidney during LN flare, potentially playing a role in renal injury.

Clinical and laboratory findings in patients with leukocyte adhesion deficiency type I: A multicenter study in Turkey

Ismail Yaz et al.

Summary: Leukocyte adhesion deficiency type I is a rare immunodeficiency disorder caused by mutations in the ITGB2 gene. Patients often have early diagnosis and consanguineous parents. Clinical features include delayed cord separation and leukocytosis. Genetic evaluation reveals various ITGB2 gene variants, with some patients undergoing hematopoietic stem cell transplantation.

Tetrandrine may treat membranous glomerulopathy via P13K/Akt signaling pathway regulation: therapeutic mechanism validation using Heymann nephritis rat model

Jiazhen Yin et al.

Summary: Membranous glomerulopathy (MGN) is an autoimmune kidney disease causing nephrotic syndrome (NS) in adults, and tetrandrine has been identified as a potential therapeutic intervention. Network pharmacology analysis and experimental validation confirmed tetrandrine's effectiveness in treating NS by targeting specific pathways and molecules. The study suggests that tetrandrine can attenuate MGN through the PI3K/Akt signaling pathway, providing new insights into its management.
