NFIA and NFIB function as tumour suppressors in high-grade glioma in mice

Kok-Siong Chen et al.

Summary: Nuclear factor one (NFI) proteins may act as tumor suppressors in glioma. Deletion of NFI genes results in reduced survival time, increased tumor load, and a more aggressive tumor phenotype in mice. NFI genes could be a credible target for both diagnostic analyses and therapeutic strategies to combat high-grade glioma.

Metabolism and function of polyamines in cancer progression

Ita Novita Sari et al.

Summary: Polyamines, essential for the proliferation and development of eukaryotes, are low-molecular-weight organic polycations with strict intracellular regulation. In cancer, dysregulation of polyamine metabolism leads to elevated levels and interactions with oncogenic pathways like mTOR and RAS, making them potential therapeutic targets. Unraveling the molecular mechanisms linking polyamine metabolism to carcinogenesis is crucial for developing novel inhibitors and understanding the findings of clinical trials.

The NFIB-ERO1A axis promotes breast cancer metastatic colonization of disseminated tumour cells

Federica Zilli et al.

Summary: NFIB is a transcription factor that plays a crucial role in promoting primary mammary tumor growth and lung metastatic colonization by increasing the expression of ERO1A. Its high expression is associated with poor prognosis in basal-like breast cancer patients.

NFIB promotes the progression of gastric cancer by upregulating circMAP7D1 to stabilize HER2 mRNA

Huimin Yang et al.

Summary: The study found that NFIB expression is increased in gastric cancer, silencing NFIB reduces cell proliferation and enhances apoptosis, circMAP7D1 may serve as a downstream molecule of NFIB affecting cell proliferation and apoptosis in gastric cancer.

Key Enzymes in Pyrimidine Synthesis, CAD and CPS1, Predict Prognosis in Hepatocellular Carcinoma

Dirk Andreas Ridder et al.

Summary: Patients with hepatocellular carcinoma have a highly variable clinical course, requiring the identification of new prognostic markers for prognosis determination and therapy selection. Dysregulated expression of key enzymes in pyrimidine synthesis was found to be associated with shorter survival in liver cancer patients. This study suggests that inhibition of pyrimidine synthesis could be a novel therapeutic strategy for HCC.

Argininosuccinate synthase 1 suppresses tumor progression through activation of PERK/eIF2α/ATF4/CHOP axis in hepatocellular carcinoma

Sanghwa Kim et al.

Summary: The expression of ASS1 was higher in HCC spheroids than in monolayer cells, and high levels of ASS1 were associated with favorable overall survival in Korean HCC patients. Bidirectional interactions between ASS1 ER stress responses in HCC-derived spheroids limit HCC progression. Overexpression of ASS1 inhibited tumor growth and enhanced the efficacy of anti-HCC chemotherapy via activation of the PERK/eIF2 alpha/ATF4/CHOP axis, independent of p53 and arginine metabolism.

NFIB promotes the migration and progression of kidney renal clear cell carcinoma by regulating PINK1 transcription

Ninghua Wang et al.

Summary: NFIB expression is significantly higher in KIRC, associated with poor prognosis, promoting progression and metastasis by regulating PINK1; NFIB can serve as a predictor and therapeutic target for KIRC.

Oncogenic KRAS creates an aspartate metabolism signature in colorectal cancer cells

Peter F. Doubleday et al.

Summary: The study highlights the significant role of KRAS mutations in colorectal cancer, particularly the upregulation of aspartate metabolizing proteins by KRAS(G13D) expression. Additionally, an increase in urea cycle enzymes expression was observed in tumor tissues. Expression of ASS1 promotes colorectal cancer cell proliferation, and high levels of various metabolites can rescue the loss of ASS1.
