Fungal iron homeostasis with a focus on Aspergillus fumigatus

Matthias Misslinger et al.

Summary: Maintaining iron homeostasis is crucial for fungi to avoid toxic excess and ensure sufficient supply. Iron plays a key role in virulence as pathogens often encounter iron limitation in host niches. Aspergillus fumigatus, a facultative human pathogen, has emerged as a model for studying fungal iron homeostasis.

The sterol C-14 reductase Erg24 is responsible for ergosterol biosynthesis and ion homeostasis in Aspergillus fumigatus

Yeqi Li et al.

Summary: The critical role of Erg24 in Aspergillus fumigatus was demonstrated in this study, showing its importance in hyphal growth, conidiation, virulence, ergosterol biosynthesis, and ion homeostasis. Loss of Erg24 resulted in accumulation of sterol intermediate and severe growth defects, which could be rescued by addition of metal ions and overexpression of ion transporters, indicating a tight link between Erg24 and ion homeostasis. These findings have important implications for antifungal discovery.

The Copper Chaperone CcsA, Coupled with Superoxide Dismutase SodA, Mediates the Oxidative Stress Response in Aspergillus fumigatus

Wenlong Du et al.

Summary: This study revealed that CcsA functions as a chaperone for SodA in A. fumigatus, and the conserved CXC motif within CcsA is crucial for its interaction with SodA and the oxidative stress response mediated by the CcsA-SodA complex. The findings provide new insights into how fungal pathogens adapt to oxidative stress via the CcsA-SodA complex.

Metal homeostasis in pathogenic Epsilonproteobacteria: mechanisms of acquisition, efflux, and regulation

Brittni R. Kelley et al.

Summary: Epsilonproteobacteria is a diverse class of eubacteria within the Proteobacteria phylum, including environmental sulfur-reducing bacteria and human pathogens that infect multiple animal hosts, causing various disease outcomes. Hosts have evolved strategies to sequester metals from pathogens or exploit metal toxicity for antimicrobial purposes, while the pathogens have developed mechanisms to sense metal availability changes and regulate their physiology in response.

Calcineurin-mediated intracellular organelle calcium homeostasis is required for the survival of fungal pathogens upon extracellular calcium stimuli

Chi Zhang et al.

Summary: Calcineurin dysfunction disrupts intracellular calcium homeostasis and leads to severe cell death, which can be significantly suppressed by alleviating cytosolic calcium overload or increasing vacuolar calcium storage capacity. This sheds light on how calcineurin regulates intracellular calcium homeostasis for cell survival and could have important implications for antifungal therapy and clinical drug administration.
