Nitric oxide protects against cochlear hair cell damage and noise-induced hearing loss through glucose metabolic reprogramming

Haidi Yang et al.

Summary: Nitric oxide (NO) plays a critical role in the pathogenesis of noise-induced hearing loss (NIHL) by modulating glucose metabolism to protect cochlear hair cells from oxidative stress and prevent NIHL.

New antioxidants from the culture broth of Coprinopsis echinospora

Dae-Won Ki et al.

Summary: Three new sesquiterpene derivatives were isolated from the culture broth of Coprinopsis echinospora during the search for natural antioxidants from fungal metabolites. Their structures were determined by spectroscopic methods, and the compounds exhibited antioxidant activity in the ABTS radical-scavenging assay with IC50 values ranging from 34.4 to 144.5 μM.

ROS-Induced Oxidative Damage and Mitochondrial Dysfunction Mediated by Inhibition of SIRT3 in Cultured Cochlear Cells

Lingjun Zhang et al.

Summary: This study investigated the damaging effects of reactive oxygen species (ROS) on sensorineural hearing loss (SNHL) and the role of sirtuin 3 (SIRT3). The results showed that ROS induced mitochondrial damage, leading to hair cell degeneration and apoptosis. SIRT3 played a crucial role in preserving mitochondrial function and protecting the cochlea from oxidative damage.

Identification of an exporter that regulates vitamin C supply from blood to the brain

Hiroshi Miyata et al.

Summary: In this study, we identified GLUT12/SLC2Al2 as a physiologically important vitamin C (VC) efflux protein expressed in the choroid plexus. Knockout of Glut12/Slc2a12 resulted in a halving of cerebral VC levels, increased VC accumulation in the choroid plexus, and reduced cerebrospinal fluid VC levels. These findings enhance our understanding of VC regulation and its physiological impact in the body.

Neuroprotective and Therapeutic Effects of Tocovid and Twendee-X on Aβ Oligomer-Induced Damage in the SH-SY5Y Cell Line

Xiao Hu et al.

Summary: In this study, the effects of two antioxidants, Tocovid and Twendee-X, on A beta O-induced damage in SH-SY5Y cells were investigated. The results showed that Tocovid and Twendee-X significantly restored cell viability, attenuated Tau protein phosphorylation, improved cell morphology, and suppressed intracellular reactive oxygen species. These findings suggest the neuroprotective and therapeutic potential of Tocovid and Twendee-X for AD treatment.

Selenoprotein P-mediated reductive stress impairs cold-induced thermogenesis in brown fat

Swe Mar Oo et al.

Summary: The study found that serum levels of selenoprotein P (SeP) are negatively correlated with brown adipose tissue (BAT) activity. Physiological cold exposure downregulates the expression of Selenop in BAT. Selenop knockout mice have higher rectal temperatures and UCP1 sulfenylation during cold exposure. Treatment with SeP in brown adipocytes reduces NA-induced mitochondrial reactive oxygen species (ROS) production by upregulating glutathione peroxidase 4, impairing cellular thermogenesis. A high-fat/high-sucrose diet increases serum SeP levels and diminishes NA-induced thermogenesis in BAT-Selenop knockout mice. Therefore, SeP acts as an intrinsic factor inducing reductive stress and impairing thermogenesis in BAT, making it a potential therapeutic target for obesity and diabetes.

Nox3-Derived Superoxide in Cochleae Induces Sensorineural Hearing Loss

Hiroaki Mohri et al.

Summary: Research has shown that the expression of Nox3 in the inner ear is associated with sensorineural hearing loss (SNHL), particularly playing a significant role in cisplatin-induced hearing loss, age-related hearing loss, and noise-induced hearing loss. This suggests that inhibiting Nox3 in the cochlea may be a promising strategy for treating ROS-related SNHL.

Antioxidant and Anti-Aging Potential of Indian Sandalwood Oil against Environmental Stressors In Vitro and Ex Vivo

Veronique Francois-Newton et al.

Summary: Indian sandalwood oil, distilled from Santalum album heartwood, has been historically used in cosmetics as a natural active ingredient with antioxidant, anti-inflammatory, and anti-proliferative activities. Studies show its superior antioxidant capacity compared to vitamin E (alpha tocopherol) in scavenging reactive oxygen species (ROS), as well as its ability to inhibit pollutant-induced collagenase levels in human skin explants, making it a potential protective and anti-aging ingredient against environmental stressors in cosmetics and dermatology.
