Full-coverage regulations of autophagy by ROS: from induction to maturation

Jing Zhou et al.

Summary: Autophagy is a well-conserved recycling process in response to stress, removing oxidized/damaged proteins and organelles to restrict further reactive oxygen species (ROS) production. Oxidative regulations occur throughout all stages of autophagy, including induction, nucleation, expansion, maturation, delivery and degradation, as well as gene transcription. Direct thiol oxidation of key proteins and certain chaperone-like proteins play roles in regulating different aspects of autophagy, while ROS can also inhibit autophagy through oxidation of key regulators.

Cellular functions of the protein kinase ATM and their relevance to human disease

Ji-Hoon Lee et al.

Summary: Deficiency in the protein kinase ATM - a master regulator of double-strand DNA breaks and stress responses - causes ataxia telangiectasia (A-T). Recent studies link A-T with other neurodegenerative disorders, and implicate reactive oxygen species, mitochondrial dysfunction, defects in proteostasis and metabolism, and increased poly(ADP-ribosyl)ation in the aetiology of A-T.

ATM at the crossroads of reactive oxygen species and autophagy

Xiaochen Xie et al.

Summary: ROS, initially considered pathological, are now recognized as signaling intermediates that promote cellular adaptation to stress through autophagy regulation. ATM protein, activated by ROS, is involved in autophagy regulation, maintaining genome stability and triggering cytoplasmic autophagy as a ROS sensor.
