New insights into the role and therapeutic potential of HSP70 in diabetes

Summary: Emerging evidence suggests that HSP70 plays a crucial role in the development of beta-cell dysfunction, insulin resistance, and diabetic complications. Hyperglycemia increases the circulating levels of HSP70, but its effects on the intracellular fraction of the protein are still unclear. It is believed that diabetic arterial structures have reduced levels of intracellular HSP70, and this systemic reduction may also occur in other systems in the body. Toll-like receptor 4 and its downstream signal transduction pathways are consistently involved in the actions of both circulating and intracellular HSP70. This review investigates the various roles of HSP70 in organs, tissues, and cells affected by hyperglycemia, with a focus on exploring pharmacological and non-pharmacological approaches targeting this protein in the context of diabetes.

Dissecting the interaction between HSP70 and vascular contraction: role of Ca2+ handling mechanisms

Summary: In this study, it was found that blockade of HSP70 leads to mishandling of Ca2+ during vascular contraction, reducing contraction amplitudes. The results also suggest that HSP70 has a positive effect on IP3r-mediated Ca2+ release and voltage-dependent Ca2+ channels. Additionally, the study indicates that HSP70 does not affect Rho-kinase or SERCA pump in the modulation of vascular contraction.

Summary: This study found differences in HSP70 expression levels between female and male rats, as well as a greater impact of HSP70 inhibition on vascular contraction in female animals. This suggests that sex may play a role in the effect of HSP70 on vascular contraction.
