The interplay of self-assembly and target binding in centrin 1 from Toxoplasma gondii

Summary: In this study, the metal binding and structural features of centrin isoform 1 in Toxoplasma gondii, TgCEN1, were characterized, revealing its properties as a Ca2+ sensor and its Ca2+-dependent self-assembly tendency. The self-assembly of TgCEN1 was found to be mediated by various physicochemical factors and can be modulated by removal of specific residues and the presence of a binding target peptide. The study also identified two distinct binding sites with different affinities and Ca2+ sensitivity on TgCEN1, suggesting a specific mechanism for Ca2+-modulated target binding.
