A genetically encoded sensor for in vivo imaging of orexin neuropeptides

Loic Duffet et al.

Summary: This study developed a genetically encoded orexin sensor called OxLight1, which enables sensitive and direct optical detection of orexin neuropeptides in living animals with high spatiotemporal resolution. The sensor was used in mouse experiments to show that orexin release is associated with behaviors such as running, stress, and sleep-to-wake transitions.

Inhibition of unfolded protein response prevents post-anesthesia neuronal hyperactivity and synapse loss in aged mice

Kai Chen et al.

Summary: Delirium is a common complication in older patients after prolonged anesthesia and surgery, and it is associated with cognitive decline and dementia. The neuronal pathogenesis of postoperative delirium is not well understood. A study in aged mice found that exposure to the anesthetic sevoflurane caused abnormal unfolded protein response (UPR), leading to neuronal hyperactivity, synapse loss, and cognitive decline. Inhibition of PERK, a marker of UPR activation, reversed these effects.

Large-scale two-photon calcium imaging in freely moving mice

Weijian Zong et al.

Summary: We developed a miniaturized two-photon microscope (MINI2P) that can perform fast and high-resolution calcium imaging of over 1,000 neurons in multiple planes simultaneously. The microscope allows stable imaging without interfering with the behavior of freely moving mice, and its optical system design improves cell yield and enables recording of a large number of neurons.

General anesthesia globally synchronizes activity selectively in layer 5 cortical pyramidal neurons

Arjun Bharioke et al.

Summary: General anesthetics induce synchronized activity in mouse layer 5 pyramidal neurons, which coincides with the loss and recovery of consciousness. This suggests that brain-wide synchrony in layer 5 pyramidal neurons may play a key role in anesthesia.

Selective optogenetic activation of orexinergic terminals in the basal forebrain and locus coeruleus promotes emergence from isoflurane anaesthesia in rats

Dan Wang et al.

Summary: The study revealed that activation of orexinergic terminals in the BF or LC facilitates emergence from anesthesia during isoflurane anesthesia, as evidenced by reduced burst suppression ratio and shorter emergence time. Additionally, optical stimulation of orexinergic terminals in the BF and LC improved rat movement scores under 0.8 vol% isoflurane anesthesia.

Dopamine D1 Receptor in the Nucleus Accumbens Modulates the Emergence from Propofol Anesthesia in Rat

Summary: Systemic activation of D1 receptors has been found to promote emergence from propofol-induced unconsciousness, with the nucleus accumbens (NAc) showing disinhibited neuronal activity during the recovery phase. D1 receptor modulation on the activity of NAc MSNs plays a vital role in facilitating the emergence from propofol-induced unconsciousness, as shown by whole-cell patch clamp recordings and pharmacological intervention experiments.

Dorsal raphe serotonergic neurons promote arousal from isoflurane anesthesia

Ao Li et al.

Summary: The dorsal raphe nucleus serotonergic neurons play a regulatory role in general anesthesia, as activation can facilitate emergence from anesthesia partly through 5-HT 1A and 2C receptors. By assessing neuronal activities, researchers found that Fos expression and calcium activity are significantly decreased during general anesthesia.

Summary: The study found that GABAergic neurons in the lateral septum play a crucial role in maintaining wakefulness and promoting emergence from anesthesia.
