In vitro preparation of uniform and nucleic acid free hepatitis B core particles through an optimized disassembly-purification-reassembly process

Yao Zhang et al.

Summary: This study developed an in vitro optimized process to prepare uniform and nucleic acid free HBc-based VLPs, successfully obtaining two types of VLPs through a disassembly-purification-reassembly procedure. It was found that the degree of disassembly had a significant impact on the protein yield, nucleic acid removal, and reassembly efficiency of the VLPs.

Production of the HBc Protein from Different HBV Genotypes in E. coli. Use of Reassociated HBc VLPs for Packaging of ss- and dsRNA

Ivars Petrovskis et al.

Summary: The core proteins (HBc) of hepatitis B virus (HBV) genotypes A, B, C, D, E, F, and G were cloned and expressed in Escherichia coli (E. coli), and the VLPs formed by HBc were purified with various methods. The best VLP yield was observed for HBV genotypes D and G, with successful packaging of RNA demonstrated.

Process development for cross-flow diafiltration-based VLP disassembly: A novel high-throughput screening approach

Nils Hillebrandt et al.

Summary: By developing a low-volume and high-resolution disassembly screening, the project provided real-time insight into the progress of VLP disassembly. Investigating two variants of C-terminally truncated hepatitis B core antigen, it was found that they exhibited different disassembly behaviors. Based on high-throughput screening results, a diafiltration-based disassembly process step was successfully developed, resulting in higher yields compared to mixing-based disassembly and allowing for integrated purification.
